Fetal thyroid tissue is a potent source of autocrine growth factor(s) that stimulate the growth of rat thyroid cells in vitro. This growth-promoting activity in fetal thyroid extract was partially purified by cation exchange and reverse-phase chromatography and was found to be distinct from insulin. Antibodies to insulin, a growth factor involved in the stimulation of 3T3L1 adipose cells and rat thyroid cells in culture, were also used to precipitate this activity, suggesting that this growth factor is not insulin. The factor(s) in fetal thyroid extract was isolated using the same three-step purification method used for insulin-like growth factor-I (IGF-I). In the second step of this purification, the factor was found to have an affinity for heparin-Sepharose, suggesting that it is a heparin-binding growth factor. Several lines of evidence suggest that the growth factor in fetal thyroid extract is IGF-I. IGF-I was present in fetal thyroid extract in high concentrations (8 ng/ml of medium), immunoprecipitated with anti-IGF-I antibodies, and was indistinguishable from recombinant IGF-I on reverse-phase high performance liquid chromatography. However, in the presence of a 100-fold molar excess of IGF-I, the thyroid extract-promoted growth of thyroid cells was only slightly inhibited. Furthermore, the thyroid extract was found to contain a previously undescribed immunoreactive IGF-I-like material that was extracted from fetal thyroid using octanoic acid and separated from IGF-I using heparin-Sepharose.
